i am doing an elisa on 96 well plate and i was wondering if i can do the following:

I want to incubate top 3 rows of assay for 30 minutes with alkaline phosphatase and then add stop solution, incubate next 3 rows for 45 minutes with alkaline Phosphatase and then add stop solution and then incubate last 2 rows for 60 minutes with alkaline phosphatase and then add stop solution.

For the top 3 rows that have only been incubated for 30 minutes, i will have to wait for 30 minutes before reading the plate (because i have to wait until bottom 2 rows have finished incubation for 60 minutes).

Will this affect the assay, or is it okay if i do this?

 Why not start with the 60

 Why not start with the 60 minute treatment, 15 minutes later start the 45 minute treatment and 30 minutes after this start the half hour treatment? That way the will all end at the same time!
