Hi! I need some help. I want to determine caspase 3 by spectrophotometer using a colorimetric kit, but I don't know if will work properly because the samples are copepods homogenates and the kit is for human caspases. Could someone help me?
The Casp-3 active site is fairly conserved between mice and humans. If the copepod (I had to look it up!) sequence is known, thats great, but if not, just set up a trial expt with a ctrl, and a known cell death agent (e.g.dexamethasone). If you only have access to homogenates, just make some mouse/human cell line lysates (-/+ dexa) as your control.
