dear fellows

how long i can store my clone culture at 4°C to be effective?

Hi
What is meant by clone culture? Is it a LB broth or other media culture of an islated clone from an agar ?
You should consider making a glycerol stock of the culture and then you need not worry about storage at 40C. Additionally you could also make a patch plate from the original isolate and store  in refrigerator.

thanks for your reply. infect i am talking about the LB broth of an isolated clone from agar plate. how long this culture can remain effective at 4C without glycerol?

Hola,quaiseruaf, I have seen recover a viable culture from a dry LB plate with the agar rigid , citting a fragment and incubating again in liquid LB. Otherwise, if all the plate is of the same clon, you can wet the surface with liquid LB and use it for a new culture. Good luck

 It is not a good idea to  use the  LB broth culture  of an isolated clone for growing the clone again as the clone could have acuumulated mutations  and ehnce non longer a pure clone. I have not used LB broth at 40C longer than 48 hrs for growing the clone again. I have seen long strands in LB broth after 48 hrs and with changes suggestive of autolysis. In case you want to gorm again the clone , you should go back to the original plate as a standard measure.
