Tool to help put silent mutations into your mutagensis primers

Just wanted to share a nice tool I use when designinig primers for site directed mutagenesis.

In addition to the desired mutation I always introduce a silent mutation that creates a unique restriction site in the mutated construct, or destroys onw in the wild type.  This helps me easily screen for clones positive for the desired mutation.

the tool I have found useful to help quickly find a silent mutation is WatCut at http://watcut.uwaterloo.ca/watcut/watcut/template.php
