Hello everyone, I have recently been examining the effects of ionizing radiation induced DNA double strand breaks in relation to the cell cycle, specifically the S-phase.
A good way to examine DNA repair is to examine the S-phase of the cell cycle using radioresistant DNA synthesis (RDS).
RDS involves tracing Brdu incorporation following ionizing radiation or chemically induced DNA damage over varying increasing doses of either IR or chemical, and using flow cytometry.
In a normal situation one would see a decrease in uptake in a functional S-phase checkpoint but in a situation where the S-phase is dysfunctional a large amount of incorporation is observed no matter how much DNA damage occurs.
