The value of Rm as detected by the test seal function of ClampEx software in whole-cell configuation is usually taken as the measure of membrane resistance. However, there is also a seal resistance in parallel with the membrane resitance. The initial giga-ohm seal between the membrane and the pipette can decrease (be partially broken) upon rupturing of the cell membrane while going whole-ell. In my understanding, this will affect measurements of Rm by ClampEx. My question is: how this potential decrease in the resistance of the seal between the membrane and the pipette is taken into account when measuring the membrane resistance?
