patch clamp and cell death

 I am doing some action potential recording for human embryonic stem cell derived cardiomyocytes, my problem is that right after I got the G-seal, my cell dead and I could not get any recording, even though it beat second before I patched the cell. We can get action potential recording from 3-5 cell cluster, also we can record the currnt from sigle cardiomycoytes. Do you think our buffer system kills the cell?

I guess I would measure the access resistance after going into the whole cell configuration. If access resistance is too low, then you have a leaky seal, which would explain the lack of action potentials.
