I was wondering if a cryopreserved tissue in sucrose/formalin could be also used for paraffin embedding, that is, going through series of alcohol after sucrose cryopreservation?
We do fat stains ie frozen sections on fixed adrenal glands. These are soaked in 7.5% sucrose for 1hr then frozen at -20c this helps cryostat sectioning. After they are processed the usual way no problems arise except occassionally crystal artifact
Thanks so much for your reply. What I meant is not processing the cryosections but the sucrose embedded tissues kept at 4 degrees for paraffin embedding thereafter.
