What is the function of phosphate balance saline in separation of blood cells ?

i think what you had mentioned here as phosphate  balance saline is phosphate buffered saline
if its so then , its a buffer which helps in   maintaining the Osmolarity and pH of cells

As Pundir has said, Phosphate Buffered Saline or PBS, is an isotonic minimal salt solution which can be used, in the context of blood cell isolation, to briefly suspend or wash the cells in before they are transferred into a more complete culture medium such as DMEM with serum where they can be maintained for a time in culture.
